Overview

Platelet aggregation is the process by which circulating platelets adhere to one another and clump together to form a plug at a site of blood vessel injury, a central early step in the formation of a blood clot. It is triggered when platelets are activated by stimuli such as exposed collagen from damaged vessel walls, thrombin, and agonists including adenosine diphosphate (ADP) and thromboxane A2. Activated platelets change shape, release their granule contents, and link to one another through fibrinogen bridges that bind to surface glycoprotein IIb/IIIa receptors. This response is essential for normal hemostasis, but excessive or inappropriate aggregation contributes to pathological thrombosis, while impaired aggregation can predispose to bleeding. Because of its dual role, platelet aggregation is a major target for antiplatelet drugs and a key measurement in clinical laboratory testing.
